On Tuesday, BioNTech SE (NASDAQ:BNTX) reported initial findings from the AHEAD-MERIT Phase 2/3 clinical trial assessing BNT113, an investigational mRNA cancer immunotherapy.
Following the announcement, the stock fell more than 5%.
The study evaluates BNT113 paired with Merck & Co. Inc.’s (NYSE:MRK) Keytruda (pembrolizumab) versus pembrolizumab alone as an initial treatment for patients suffering from advanced head and neck cancer.
Specifically, the study focuses on individuals with unresectable recurrent or metastatic human papillomavirus 16-positive (HPV16+) head and neck squamous cell carcinoma (HNSCC) exhibiting PD-L1.
Progression-Free Survival Endpoint Missed
The late-stage portion of the study utilizes complementary dual primary endpoints: progression-free survival (PFS) and overall survival (OS).
During the initial analysis, the combination of BNT113 and pembrolizumab did not achieve a statistically significant improvement in progression-free survival when compared directly to the pembrolizumab monotherapy.
BioNTech said that even with this setback, mRNA cancer immunotherapies continue to serve as a foundational element in the oncology strategy for the company.
The broader pipeline prioritizes innovative treatment combinations designed to elevate patient outcomes.
Focus Shifts To Overall Survival
Following these initial results, the Independent Data Monitoring Committee advised that the trial proceed as originally planned.
The focus now shifts toward evaluating the second primary endpoint of overall survival to determine if the combination therapy can help patients live longer.
According to the predetermined statistical framework of the study, securing a statistically significant enhancement in overall survival will classify the trial as a success, even though the progression-free survival target went unmet.
The company currently remains blinded to the active trial data.
Addressing Unmet Medical Needs
Medical providers currently have no approved therapies specifically targeting HPV-positive tumors. HPV infections link to roughly one-third of all HNSCC cases, with the high-risk HPV16 variant driving the majority of these instances.
Because many patients experience disease progression while receiving standard-of-care treatments, the medical community needs novel options that improve long-term survival rates.
BNT113 works by encoding a fixed antigen set to explicitly target two HPV16-derived antigens frequently present on these specific tumor cells.
BNTX Price Action: BioNTech shares were down 5.22% at $90.84 at the time of publication on Tuesday, according to Benzinga Pro data.
