SENTI-202 is designed to selectively target and eliminate CD33 and/or FLT3-expressing hematologic malignancies while sparing healthy bone marrow cells.
Three AML patients have been treated at the lowest dose level (1.0 billion CAR+ NK cells per dose) and, as of the data cutoff date of September 19, two achieved complete remission, confirmed by bone marrow biopsy, which includes blast reduction and recovery of blood cells to normal ranges.
In addition, both patients were assessed as measurable residual disease negative after treatment, defined as no detectable cancer cells in a bone marrow sample.
Both patients maintain remission (4+ months and 3+ months, respectively).
The Safety Review Committee cleared the lowest dose cohort, and dose escalation continues at the 1.5 billion CAR+ NK cells/dose level.
SENTI-202 was generally well-tolerated with no dose-limiting toxicities and an adverse event profile consistent with other investigational NK cell therapies and patients with underlying AML receiving lymphodepleting chemotherapy.
SENTI-202 transgene was detected in the peripheral circulation of all three patients and all cycles, with a pharmacokinetic profile generally consistent with other investigational CAR-NK therapy levels.
The company expects to enroll approximately 20 patients in the Phase 1 trial. A higher dose cohort of 1.5 billion CAR+ NK cells/dose is actively enrolling.
Additional safety and efficacy data, including initial durability data, are expected in 2025.
Concurrently, Senti Biosciences announced a private investment in public equity financing to issue and sell an aggregate of 16,713 shares of Series A Convertible Preferred Stock, with gross proceeds of <a target="_blank" href="https://www.benzinga.com/pressreleases/24/12/g42263313/senti-biosciences-inc-announces-oversubscribed-37-6-million-private-placement-equity-financing" rel="noreferrer noopener">approximately $37.6 million</a>.
The company has also granted a certain investor an option to purchase an additional 4,444 shares of Preferred Stock and accompanying warrants for gross proceeds of approximately $10.0 million.
Senti Biosciences also announced that it has appointed Fran Schulz, an experienced biotechnology and financial executive, to the Senti Bio Board of Directors. Omid Farokhzad is departing from the <a target="_blank" href="https://www.benzinga.com//node/42262836" rel="noreferrer noopener">Board in conjunction with Schulz&#8217;s appointment.</a>

Last week, Walmart Corp. (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/WMT#NYSE">WMT</a>) became the latest major corporation to roll back its diversity, equity and inclusion — DEI — policies. Walmart joins other companies like Tractor Supply Co. (NASDAQ:<a class="ticker" href="https://www.benzinga.com/stock/TSCO#NASDAQ">TSCO</a>) and Lowe's Co. (NYSE:<a class="ticker" href="https://www.benzinga.com/stock/LOW#NYSE">LOW</a>), all of which ended their participation in Human Rights Campaign surveys and removed supplier diversity spending goals. 
Fishback estimates the Meritocracy fund's criteria will eliminate approximately two dozen companies from being eligible for inclusion in the fund. He also intends to pressure excluded companies to drop their DEI policies through public campaigns and behind-the-scenes engagement.&nbsp;
According to <a href="https://www.semafor.com/article/11/26/2024/the-sp-500-without-the-woke-sht-here-come-the-maga-funds">a report</a> from Semafor, Azoria is finalizing a roughly $25 million venture round that Fishback said is sourced from "two prominent conservative-minded California venture capitalists" and family offices.&nbsp;
The firm is also planning more funds in the future. One of the planned future ETFs would trade under the ticker "BTFD" and would "buy the dip" when stocks fall sharply.&nbsp;

The U.S. pharmaceutical industry is actively lobbying for revisions to the Medicare drug price negotiation provisions of the Inflation Reduction Act (IRA).
The IRA empowers Medicare to negotiate prices for costly drugs, potentially leading to price cuts ranging from the statutory minimum&nbsp;<a target="_blank" href="https://www.benzinga.com/government/24/01/36849742/medicares-historic-negotiations-to-lower-drug-prices-analysts-anticipate-upto-potential-price-cuts-o" rel="noreferrer noopener">of 25% to a significant 60%</a>.&nbsp;
In September, the U.S. Department of Health and Human Services (HHS), working via the Centers for Medicare &amp; Medicaid Services (CMS), released final guidance for the second cycle of the&nbsp;<a target="_blank" href="https://www.benzinga.com/general/health-care" rel="noreferrer noopener">Medicare Drug Price Negotiation Program</a>.
The initiative&nbsp;aims&nbsp;to provide lower drug prices starting in 2026 and 2027.
The first round of negotiations focused on <a target="_blank" href="https://www.benzinga.com/general/biotech/24/10/41166018/medicare-drug-price-negotiation-program-gets-final-guidance-for-second-cycle-1-5b-savings-expecte" rel="noreferrer noopener">ten specific drugs</a>.
Medicare recipients could save approximately $1.5 billion in out-of-pocket costs when the new prices take effect in 2026.
Industry representatives are targeting changes under a potential Trump administration, aiming to extend the timeline for certain drugs&#8217; eligibility for negotiations, Reuters highlighted, citing sources familiar with the matter.

By February 2025, CMS will select up to 15 additional drugs for the second round of negotiations. The selected drugs will undergo negotiations in 2025, with new prices becoming effective in 2027.
President Joe Biden wants to expand the number of prescription drugs eligible for Medicare price negotiations. Instead of the current limit of 20 drugs per year, he suggests raising it to at least 50. Also, some popular weight loss and diabetes drugs could be targeted in the next round of negotiations, <a target="_blank" href="https://www.benzinga.com/general/biotech/24/08/40534249/novo-nordisks-ozempic-might-be-among-2027-medicare-price-negotiation-targets" rel="noreferrer noopener">which is set for 2027</a>.
A Reuters report highlights that lobbyists and executives from major pharmaceutical and biotech companies are advocating for a four-year delay in negotiating eligibility for small-molecule drugs, such as pills, which comprise the majority of medications.
Under current rules, these drugs are subject to negotiations nine years after launch, compared to 13 years for biologics. Drugmakers argue this timeline discourages innovation for more affordable, convenient therapies.
Since its passage in 2022, the IRA has drawn industry criticism, with claims that it could stifle research and development.
Also, major pharmaceutical companies involved in the first U.S. negotiations over Medicare drug prices expect limited business impact despite&nbsp;<a target="_blank" href="https://www.benzinga.com/general/biotech/24/07/40047654/bristol-myers-johnson-johnson-abbvie-astrazeneca-expect-minimal-impact-from-upcoming-medicare-pri" rel="noreferrer noopener">anticipated price cuts</a>.
Some pharmaceutical firms have engaged directly with President-elect Donald Trump&#8217;s transition team members to propose amendments to the IRA.
These companies hope that Republican majorities in Congress will be receptive to aligning negotiation timelines for all drugs, removing distinctions between pills and biologics.
Not everyone agrees with the industry&#8217;s position. The report cites S. Sean Tu, a law professor at West Virginia University, who says the proposed changes unfairly benefit drug companies. He told Reuters that the current nine-year exclusivity period already supports innovation and warns that extending it would burden patients more without offering significant benefits.
